I've wonderd too, had she and 'Splinter' ended up in NYC together instead. I'm curious if she'd have remined human or, if more likely for story purposes if she was present, if she'd have mutated and what would she be. To think not only would the Turtles have had a mom, but she might have ended turtle herself...
I've always wanted Tang Shen to know the Turtles in the current series. Like this:

Well, after reading everyone's amazing little dialouges (they were awesome everyone ;) be proud of yourselves!) I figured what the heck, I'll take a shot at it. I think I'll just write out the inital meeting scene, I have no little backstories on how she came to be not dead and such. Alright, here goes...

(April is sitting in her room, tapping her foot nervously. Her eyes dart around the room and she holds a phone up to her ear. It rings once...twice...then the line suddenly clicks and a voice leaks through)
Donatello: (from other end of phone) April?
April: Hey Donnie.
Donatello: Hi...so....um....
April: It's urgent Donnie. (glances at door) Something...I don't even know what's going on. I'm bringing someone down to the lair. If she is who she claims to be...(shakes head) What happens to people when they go into shock?
Donatello: Shock? What's going on April?
April: Splinter may go into shock, alright? Just figured I'd warn you.
(April quickly hangs up the phone, then exhales deeply, running her fingers through her hair)
April: Dear Lord. Here goes nothing.
*scene switches to the sewer lair*
(April walks in, passing through the turnstiles. A middle-aged Japanese woman is following her. She has long black hair and is somewhat attractive despite her age. Her eyes dart around the lair curiously and she plays with her hands)
Tang Shen: ...So where are we again....what was your name?....April, right. Where are we April?
April: I'm taking you to see Hamato Yoshi.
(Tang Shen frowns and looks around her, raising an eyebrow somewhat doubtfully, but makes no comment)
April: Splinter! Guys! (voice cracks) There's someone here to see you...
(Leonardo emerges cautiously from the kitchen, his eyes flickering between April and Tang Shen)
Leonardo: Who is that?
(Tang Shen takes a step backwards somewhat fearfully)
Tang Shen: What is that?
April: Leo! (quickly ushers him back into the kitchen) I think Splinter should see her first...
Leonardo: (frowns) What the heck is going on April?
(As April shoves Leonardo back into the kitchen, Splinter comes out of the dojo, his eyes narrowing as he looks at April)
Splinter: April? What is...
(Splinter's eyes wander and suddenly focus on Tang Shen. He takes in a sharp breath, his muscles tensing and his eyes widening. Tang Shen takes a few more steps backwards, her eyes darting from Splinter to Leo)
Tang Shen: You told me you were taking me to see Yoshi. What are these things?
(Splinter continues to stare at Tang Shen, unmoving. He makes a small noise of disbelief and his staff clatters to the floor)
April: Leo, get Donnie, I don't know what will happen to Splinter.
(Leo looks at Splinter, then back to April. Finally he stiffly walks over to the lab, disappearing into the large room)
Splinter: Tang....Shen?
(Splinter's hands are shaking now and his breaths are quick and shallow)
Tang Shen: (frowns) How does it know my name...?
(Tang Shen suddenly stares at the crest on Splinter's robes, her face one of pure shock)
Tang Shen: Why is it wearing my husband's crest?
(April quickly walks over to Tang Shen, putting her hands on the woman's shoulders)
April: Tang Shen...this is Hamato Yoshi.
(Tang Shen looks Splinter up and down, her lips moving but no noise coming out. Finally, she gathers her composure)
Tang Shen: (in a high pitched, squeaky voice) Yoshi? But how could this be him?
(Splinter suddenly begins to walk over to Tang Shen, his movements unsure and his eyes still wide in shock. Tang Shen starts to back away from him, but April quickly stops her)
April: (in a soft voice) Wait.
(Tang Shen looks at April fearfully, but remains still as Splinter approaches. Splinter looks down at Tang Shen, blinking once or twice.)
Splinter: (in a whisper) My Tang Shen....you are alive.
(Tang Shen's eyebrows furrow as she stares into Splinter's eyes. As she looks deeply into his eyes, her own slowly start to widen)
Tang Shen: It's not possible...but...
Splinter: I am Hamato Yoshi. Your Hamato Yoshi.
(tears begin to well up in Splinter's eyes as Tang Shen takes in a sharp breath, tears running down her own face)
Tang Shen: It's you. It's really you. After all these years...
(Donnie, Raph, Mikey and Leo come out of the lab and linger around the edges of the room, sharing nervous glances)
Raph: Who the heck is that?
(Splinter and Tang Shen remain frozen in place, completley absorbed in each other. April quickly walks over to the turtles)
April: Guys....It's Tang Shen.
(All the brothers eyes widen)
Donatello: But...that's not possible. She was dead. Splinter held her body in his arms.
Raph: No. That can't be her. She's dead. (shakes head in disbelief)
Leonardo: How could she possibly be...?
(Mikey splits off from the group and approaches Tang Shen. Everyone is too shocked to try to stop him. The room is silent as Mikey stops a few feet away from Tang Shen, looking up at her with wide, blue innocent eyes.)
Mikey:....Mom?
(Tang Shen blinks a few times, glancing from Splinter to Michelangelo. Suddenly her eyes brighten and she realizes that the mutant turtle is Splinter's adopted son. She looks down at Mikey, whose eyes are somewhat desperate, filled with cautious hope, and a small smile escapes her lips)
Tang Shen: I am Tang Shen, young one. If you are a son of Hamato Yoshi, you are a son of mine.
Michelangelo: (smiles)....Mom!
(Mikey suddenly engulfs Tang Shen in a hug. At first she looks panicked, but in a moment she's regained herself and she gently puts her arms around the young orange-masked turtle. She beckons Splinter over, and Splinter laughs in disbelief, joining the group hug. In a moment the other three turtles have ran over and are hugging Tang Shen as well. April lingers off the the side, a smile curling her lips)
Tang Shen: (looks at April) April O'Neil, I cannot thank you enough. You have reuinited me with my beloved Yoshi. (beckons her over) Come.
(April smiles and is quickly pulled into the hug as well. The seven of them remain silent, their eyes closed and each person's expression the very definition of happiness...)

And yes, I considered how Splinter may have wished Tang Shen to be a mother in his sons' lives. But as I analyzed the cause and effect of it all, I realized that Shen's death and Miwa's disappearance were the catalyst for Hamato Yoshi traveling to Japan, and buying four pet turtles out of loneliness. Quite simply, if Yoshi hadn't lost his first family, he wouldn't have gotten a second one. So, unless Shen can be brought back 15 years after she died, it's unlikely she could have ever been a part of the turtles' lives. I'm sure Splinter has thought this over many times. He of all people must be familiar with the mantra "You can't always get what you want."
As for the idea that the turtles are "missing a mother," I don't really think this is the case. Splinter misses Shen personally, sure. But she was never directly in the turtles' lives to begin with, so she's not missing from their lives. The truth is, Splinter was a good father, and raised four good sons, and did it largely on his own, such that he is their only parent of functional significance. I think it would be nice if they could meet Shen, but it would be harder for her to suddenly become a functional mother to them when they're already all almost 16. They're past needing a mommy to kiss their boo boos or to hug them when they're sad—that was all Splinter's job. If Shen were introduced into their lives, they would be as much a stranger to her, as she is to them. She could become their friend, and that's still nice. The paradox is, even if Splinter himself tomorrow remarries a different woman who the turtles are more familiar with, she wouldn't necessarily become the mother of four sons who are already all but independent.
The closest thing the turtles were ever going to find to a mother figure, was a big sister figure—April. This version of April isn't much older than they are (only two years older), but then, if I recall correctly, Mirage April was only four years older than the turtles, and IDW April has a similar age difference. :) In most incarnations of TMNT, April is a good stabilizing influence in their family life. But, again, she's more their sister, not their mother.
Even Karai is not functionally missing Shen—she was raised to miss a woman in a photograph, but she never got to know Shen, and grew up without Shen's help, even though the one who actually raised Karai was the same person who killed her mother. Karai is not even functionally missing Splinter, either; she's glad to have found him and they had some hugs and happy moments, and Karai even betrayed her own "father" to be with him again, but I think that more demonstrates how evil and manipulative and treacherous Oroku Saki was to begin with. As Karai demonstrates, Splinter no longer had much power to shape her, and when push came to shove, she tried to take revenge against the Shredder according to morals the Shredder himself taught her—which was exactly why the Shredder anticipated her arrival. Again, Karai and Splinter are perfectly capable of becoming friends, and Asian filial piety will compel Karai to honor and respect him as her titular father, but but it's too late for him to be her dad. It's no fault of his, or of hers—it just cannot be helped, period. Can they all become a family again? Sure, it's possible. But they would be a family of people who mainly only know each other as adults. The Shredder made sure of that.
I've been thinking. Since he found out about his Foot heritage, Shredder would've killed only Splinter and take Tang Shen and Miwa away with him thinking they could rule the Foot Clan together even though Tang Shen would refuse to go through with it and try to escape with her daughter.

I'm sure he tried. I think Shen died by accident, didn't she? And yes, Oroku Saki is possessive and cares more about his own desires and nothing for the needs of those around him except in how it serves his desires. He'll betray anyone and everyone in the end.
