Big Mama's train is dark purple and the head of the train is loosely modeled after Big Mama with eight glowing red eyes on each side, light grey teeth-like formations, two chimneys aligned in a column with the taller of the two in front, and a red and purple cowcatcher. It has dark purple wheels with seafoam green rims, a gold train whistle, and six cars with eight green windows each. Passengers board at the end car where lies a baggage car for the passengers' belongings. There is a freezer car for meat, a gym car, a theater car, a diner car, a spa car, and a miscellaneous front car. The train is mystic and the baggage car is the only car that has a conventional sliding door as rest of the cars are accessed via a wheel of 70 teleportation buttons at the end of each car. The black button accesses the front train car.
Big Mama's train travels both through the Hidden City and New York. During its New York travels, it is shielded from sight by a mystic shimmer, however, its affect on waterways remains visible. The train doesn't service human beings. Big Mama has staff stationed in the diner car and spa car to cater to passengers.

Big Mama's train debuted in "Mystic Library" in the form of a children's toy model, which Michelangelo was delighted by.
Big Mama's train reappeared in "Insane in the Mama Train". Sometime prior to the episode, the Foot Clan and Big Mama arranged a deal that she would lend her train's services and respect the privateness of their mission in exchange for ownership of the Mystic Orb. The Foot soldiers loaded the packaged Kuroi Yōroi onto the train where it was reassembled in the train's front car. April O'Neil found herself stuck on the train disguised as a Foot soldier as the train left its station for the Shredder's shrine. Later, the Turtles trespassed onto the train from its roof with Splinter's help with the intent of stealing the Kuroi Yōroi from the Foot. Mikey and April incapacitated Foot Brute in the baggage car and then swiftly moved through the diner car. Finding themselves stumped by the car's strange, mystic doorway, Mikey and Donatello separated from the team on Raphael's order to locate the Kuroi Yōroi. They were eventually confronted by Foot Lieutenant and Foot Recruit in the gym car. Leonardo and April teleported to the freezer car, but encountered one of Baron Draxum's guardsmen. As Raph finally selected a button to press, he was challenged by the other guardsmen who had been receiving a massage in the spa car. Skirmishes between the Mad Dogs and their foes ensued and Foot Recruit was teleported to the spa car by Mikey and the guardsman was teleported from the spa car to the freezer car. April and Leo struggled to combat the newfound pair of guardsmen and both Leo and Raph were compelled to teleport to the theater car where a Yōkai passenger hosted a talk involving mystic taxes. Together, they returned to the freezer car to defeat the guardsmen and teleport themselves and April to the gym car. They reunited with Mikey and Donnie and abducted Foot Lieutenant along with the guardsmen from the freezer car, Foot Recruit from the spa car, and Foot Brute from the baggage car. In the theater car, Raph restrained them all and teleported with the Mad Dogs to the train's front car. Foot Lieutenant and Foot Recruit were soon after tossed off of the train by a fellow passenger. Upon reaching the Kuroi Yōroi, the Mad Dogs were attacked by Draxum. He bound April and teleported her back to New York and kidnapped the Turtles, causing their mission to fail. The train then reached the Shredder's shrine where the Foot got off with the armor.
The toy model of Big Mama's train appeared in the uncut version of "Hidden City's Most Wanted" as one of Heather's belongings, which she tried to run Draxum over with.
